ClinVar contains an entry for this variant (Variation ID: 906393). This variant has not been reported in the literature in individuals affected with HSPB3-related conditions. This variant is present in population databases (rs755999042, gnomAD 0.005%). This sequence change creates a premature translational stop signal (p.Arg23*) in the HSPB3 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 128 amino acid(s) of the HSPB3 protein. -
This variant was observed in the ICSL laboratory as part of a predisposition screen in an ostensibly healthy population. It had not been previously curated by ICSL or reported in the Human Gene Mutation Database (HGMD: prior to June 1st, 2018), and was therefore a candidate for classification through an automated scoring system. Utilizing variant allele frequency, disease prevalence and penetrance estimates, and inheritance mode, an automated score was calculated to assess if this variant is too frequent to cause the disease.
