Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4.4e-05 in 248924 control chromosomes (gnomAD). This frequency is not significantly higher than estimated for a pathogenic variant in ADGRV1 causing Usher Syndrome (4.4e-05 vs 0.0054), allowing no conclusion about variant significance. c.3974C>T has been reported in the literature as a non-informative genotype (second allele/genotype not specified) in at-least one individual affected with features of Usher Syndrome (example, Krawitz_2014). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 25333064). Five clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 and classified the variant as VUS (n=3) and likely pathogenic (n=1).
