The p.Gly5223Arg variant in ADGRV1 (also known as GPR98) gene has not been previously reported in individuals with hearing loss or Usher syndrome and was absent from large population studies. Computational prediction tools and conservation analysis do not provide strong support for or against an impact to the protein. In summary, the clinical significance of the Gly5223Arg variant is uncertain. ACMG/AMP criteria applied: PM2. -

The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and arginine. This variant is not present in population databases (ExAC no frequency).
