This sequence change replaces arginine, which is basic and polar, with histidine, which is basic and polar, at codon 47 of the GCM2 protein (p.Arg47His). This variant is present in population databases (rs104893959, gnomAD 0.01%). This variant has not been reported in the literature in individuals affected with GCM2-related conditions. Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GCM2 protein function with a positive predictive value of 80%. This variant disrupts the p.Arg47 amino acid residue in GCM2. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 15863676, 31671402; internal data). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
