The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. This sequence change replaces isoleucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 1353 of the LAMA4 protein (p.Ile1353Val). ClinVar contains an entry for this variant (Variation ID: 541222). This variant has not been reported in the literature in individuals affected with LAMA4-related conditions. This variant is not present in population databases (gnomAD no frequency).

The p.I1353V variant (also known as c.4057A>G), located in coding exon 29 of the LAMA4 gene, results from an A to G substitution at nucleotide position 4057. The isoleucine at codon 1353 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
