Variant summary: SLC17A5 c.1355_1356insAA (p.Val453MetfsX50) causes a frameshift which results in an extension of the protein. The variant allele was found at a frequency of 8e-06 in 250158 control chromosomes (gnomAD). c.1355_1356insAA has been reported in the literature in one individual affected with Infantile Sialic Acid Storage Disorder (Aula_2000). These data indicate that the variant is very likely to be associated with disease. One ClinVar submitter (evaluation after 2014) cites the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as likely pathogenic. -
