Variant summary: COL12A1 c.8548A>G (p.Met2850Val) results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be tolerated. The variant allele was found at a frequency of 6e-05 in 249426 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in COL12A1 causing Ullrich congenital muscular dystrophy 2 (6e-05 vs 0.0035), allowing no conclusion about variant significance. To our knowledge, no occurrence of c.8548A>G in individuals affected with Ullrich congenital muscular dystrophy 2 and no experimental evidence demonstrating its impact on protein function have been reported.
