This alteration results from a G to A substitution at nucleotide position 643, causing the glutamic acid (E) at amino acid position 215 to be replaced by a lysine (K). Based on data from gnomAD, the A allele has an overall frequency of 0.007% (19/282798) total alleles studied. The highest observed frequency was 0.058% (6/10366) of Ashkenazi Jewish alleles. This alteration has been reported in a dilated cardiomyopathy cohort and an idiopathic ventricular tachycardia cohort; however, clinical details were limited in both cases and additional alterations in other cardiac-related genes were identified in one case (Verdonschot, 2020; Guelly, 2021). This amino acid position is highly conserved in available vertebrate species. The in silico prediction for this alteration is inconclusive.
