Variant summary
Our verdict is Uncertain significance. The variant received 4 ACMG points: 4P and 0B. PP3_Strong
The NM_004999.4(MYO6):c.2507G>A(p.Arg836His) variant causes a missense, splice region change involving the alteration of a conserved nucleotide. The variant allele was found at a frequency of 0.00000415 in 1,446,058 control chromosomes in the GnomAD database, with no homozygous occurrence. 3/3 splice prediction tools predicting alterations to normal splicing. Variant has been reported in ClinVar as Uncertain significance (★). Synonymous variant affecting the same amino acid position (i.e. R836R) has been classified as Uncertain significance.

Variant classified as Uncertain Significance - Favor Pathogenic. The c.2507G>A ( p.Arg836His) variant in MYO6 has not been previously reported in individuals wit h hearing loss and was absent from large population studies. This variant is loc ated at the last base of exon 24, which is a conserved nucleotide within the 5' splice site consensus region. Computational tools suggest a possible impact to s plicing; however, this information is not predictive enough to determine pathoge nicity. Additional computational prediction tools and conservation analyses do n ot provide strong support for or against an impact to the protein. In summary, w hile there is some suspicion for a pathogenic role, the clinical significance of this variant is uncertain. -
