Variant summary
Our verdict is Uncertain significance. The variant received 2 ACMG points: 2P and 0B. PM2
The NM_015021.3(ZNF292):c.406G>A(p.Ala136Thr) variant causes a missense change. The variant was absent in control chromosomes in GnomAD project. Variant has been reported in ClinVar as Uncertain significance (★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. A136V) has been classified as Uncertain significance.

Intellectual developmental disorder, autosomal dominant 64 Uncertain:1
The missense variant in c.406G>A (p.Ala136Thr) in ZNF292 gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The p.Ala136Thr variant is novel (not in any individuals) in gnomAD Exomes and 1000 Genomes. The amino acid Ala at position 136 is changed to a Thr changing protein sequence and it might alter its composition and physico-chemical properties. For these reasons, this variant has been classified as Uncertain Significance. -
