This variant is present in population databases (rs397508467, gnomAD 0.002%). This missense change has been observed in individuals with cystic fibrosis (PMID: 11732487, 14872121). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt CFTR protein function with a positive predictive value of 80%. Experimental studies have shown that this missense change affects CFTR function (PMID: 8663008, 29805046). The variant allele was found at a frequency of 8e-06 in 251082 control chromosomes (gnomAD). c.296C>T has been observed in multiple individuals affected with Cystic Fibrosis (e.g. Amaral_2001, Gilljam_2004, McCague_2019). These data indicate that the variant is very likely to be associated with disease. At least one publication reports experimental evidence evaluating an impact on protein function. The most pronounced variant effect resulted in approximately 3.9% of normal chloride channel conductance relative to wild type (e.g., Bihler_2024).
