Variant summary
Our verdict is Uncertain significance. Variant got 5 ACMG points: 6P and 1B. PM1PM2PM5BP4
The NM_000492.4(CFTR):c.1079C>T(p.Thr360Ile) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.0000316 in 1,613,676 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. 12/21 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. T360K) has been classified as Pathogenic.

Uncertain significance, criteria provided, single submitter | clinical testing | Ambry Genetics | Aug 06, 2024 | The p.T360I variant (also known as c.1079C>T), located in coding exon 8 of the CFTR gene, results from a C to T substitution at nucleotide position 1079. The threonine at codon 360 is replaced by isoleucine, an amino acid with similar properties. This variant was reported in a cohort of 105 Russian patients with chronic pancreatitis (Litvinova MM et al. Sovrem Tekhnologii Med, 2023 Mar;15:60-70). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. - |
Uncertain significance, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Oct 04, 2021 | This sequence change replaces threonine with isoleucine at codon 360 of the CFTR protein (p.Thr360Ile). The threonine residue is weakly conserved and there is a moderate physicochemical difference between threonine and isoleucine. This variant is present in population databases (rs75053309, ExAC 0.03%). This variant has not been reported in the literature in individuals affected with CFTR-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The isoleucine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. - |
