Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes a 5 splicing donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 249234 control chromosomes (gnomAD). c.1766+2T>C has been reported in the literature in compound heterozygous individuals affected with Cystic Fibrosis (Choe_2009, Maclean_2011, Petrova_2019, Erdoan_2021). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Two ClinVar submitters (evaluation after 2014) cite this variant pathogenic and likely pathogenic. Based on the evidence outlined above, the variant was classified as pathogenic. - |
