The CFTR c.3139+8A>G variant (rs193922517) is reported in the literature in an individual with borderline sweat chloride levels (Schrijver 2005). This variant is reported in ClinVar (Variation ID: 35863) and is absent from the Genome Aggregation Database, indicating it is not a common polymorphism. This is an intronic variant in a weakly conserved nucleotide, but computational analyses (Alamut v.2.11) predict that this variant may impact splicing by creating a novel cryptic donor splice, however these predictors are not considered high quality. However, given the lack of clinical and functional data, the significance of the c.3139+8A>G variant is uncertain at this time. References: Schrijver I et al. Variant summary: CFTR c.3139+8A>G alters a non-conserved nucleotide located at a position not widely known to affect splicing. Consensus agreement among computation tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 250618 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.3139+8A>G has been observed in two individuals affected with Cystic Fibrosis and suspicion of cystic fibrosis, without strong evidence for causality (Schrijver_2005_b, Pepermans_2016). These report(s) do not provide unequivocal conclusions about association of the variant with Cystic Fibrosis. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 26500004, 15858154). ClinVar contains an entry for this variant (Variation ID: 35863). Based on the evidence outlined above, the variant was classified as likely benign. -
