This sequence change creates a premature translational stop signal (p.Val249Glufs*9) in the NT5C3A gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 49 amino acid(s) of the NT5C3A protein. This variant is present in population databases (rs397518436, gnomAD 0.004%). This premature translational stop signal has been observed in individuals with pyrimidine 5'-nucleotidase deficiency (PMID: 12714505, 12930399). It has also been observed to segregate with disease in related individuals. This variant is also known as 743-744insGG and InsGG743. ClinVar contains an entry for this variant (Variation ID: 4482). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic. -
