Genes affected
MPLKIP (HGNC:16002): (M-phase specific PLK1 interacting protein) The protein encoded by this gene localizes to the centrosome during mitosis and to the midbody during cytokinesis. The protein is phosphorylated by cyclin-dependent kinase 1 during mitosis and subsequently interacts with polo-like kinase 1. The protein is thought to function in regulating mitosis and cytokinesis. Mutations in this gene result in nonphotosensitive trichothiodystrophy. [provided by RefSeq, Nov 2009]

Uncertain significance, criteria provided, single submitterclinical testingInvitaeOct 18, 2023This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 166 of the MPLKIP protein (p.Asn166Ser). This variant is present in population databases (rs557616399, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with MPLKIP-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Not Available"; PolyPhen-2: "Benign"; Align-GVGD: "Not Available". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -
