The p.D393H variant (also known as c.1177G>C), located in coding exon 10 of the EGFR gene, results from a G to C substitution at nucleotide position 1177. The aspartic acid at codon 393 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
EGFR-related lung cancer Uncertain:1
This sequence change replaces aspartic acid, which is acidic and polar, with histidine, which is basic and polar, at codon 393 of the EGFR protein (p.Asp393His). This variant is present in population databases (rs587778246, gnomAD 0.006%). This variant has not been reported in the literature in individuals affected with EGFR-related conditions. ClinVar contains an entry for this variant (Variation ID: 134018).
