ClinVar contains an entry for this variant (Variation ID: 826188). This variant is present in population databases (rs746910555, gnomAD 0.02%). This sequence change replaces asparagine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 204 of the PMS2 protein (p.Asn204Asp). -
Hereditary cancer-predisposing syndrome Uncertain:1
The p.N204D variant (also known as c.610A>G), located in coding exon 6 of the PMS2 gene, results from an A to G substitution at nucleotide position 610. The asparagine at codon 204 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear. -
