Variant summary
Our verdict is Uncertain significance. The variant received 0 ACMG points: 2P and 2B. PM2BP4_Moderate
The NM_001130965.3(SUN1):c.1582G>C(p.Gly528Arg) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.00000657 in 152,300 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. 13/20 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★★). Another variant affecting the same amino acid position, but resulting in a different missense (i.e. G528C) has been classified as Uncertain significance.

The c.1582G>C (p.G528R) alteration is located in exon 14 (coding exon 14) of the SUN1 gene. This alteration results from a G to C substitution at nucleotide position 1582, causing the glycine (G) at amino acid position 528 to be replaced by an arginine (R). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
Emery-Dreifuss muscular dystrophy Uncertain:1
In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0". The arginine amino acid residue is found in multiple mammalian species, suggesting that this missense change does not adversely affect protein function. These predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This variant has not been reported in the literature in individuals with SUN1-related conditions. This variant is present in population databases (rs549897796, ExAC 0.01%). This sequence change replaces glycine with arginine at codon 528 of the SUN1 protein (p.Gly528Arg). The glycine residue is moderately conserved and there is a moderate physicochemical difference between glycine and arginine. -
