ABCB4 (HGNC:45): (ATP binding cassette subfamily B member 4) The membrane-associated protein encoded by this gene is a member of the superfamily of ATP-binding cassette (ABC) transporters. ABC proteins transport various molecules across extra- and intra-cellular membranes. ABC genes are divided into seven distinct subfamilies (ABC1, MDR/TAP, MRP, ALD, OABP, GCN20, White). This protein is a member of the MDR/TAP subfamily. Members of the MDR/TAP subfamily are involved in multidrug resistance as well as antigen presentation. This gene encodes a full transporter and member of the p-glycoprotein family of membrane proteins with phosphatidylcholine as its substrate. The function of this protein has not yet been determined; however, it may involve transport of phospholipids from liver hepatocytes into bile. To our knowledge, this variant has not been reported in the literature. This variant is reported in 0.0062% of alleles in individuals of African descent in gnomAD. Other nucleotide substitutions leading to a similar protein extension effect (p.*1280Argext*17) has been reported in multiple individuals with progressive familial intrahepatic cholestasis (Chinnaratha et al. 2013. PubMed ID: 23574360; Hakim et al. 2019. PubMed ID: 31000363; Table1, Bakır et al. 2021. PubMed ID: 34961929). Although we suspect this variant may be pathogenic, at this time, the clinical significance of the c.3839G>C (p.*1280Serext*19) variant is uncertain due to the absence of conclusive functional and genetic evidence. - |
