Long QT syndrome Uncertain:1
This variant is present in population databases (rs755140795, ExAC 0.005%) but has not been reported in the literature in individuals with a AKAP9-related disease. This sequence change replaces serine with cystine at codon 144 of the AKAP9 protein (p.Ser144Cys). The serine residue is highly conserved and there is a moderate physicochemical difference between serine and cystine. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "deleterious", MutationTaster: "polymorphism", Align-GVGD: "Class C65"). In summary, this variant is a rare missense change with uncertain impact on protein function. There is no indication that it causes disease, but the available evidence is currently insufficient to prove that conclusively. Therefore, it has been classified as a Variant of Uncertain Significance. -
Cardiovascular phenotype Uncertain:1
The c.431C>G (p.S144C) alteration is located in exon 5 (coding exon 5) of the AKAP9 gene. This alteration results from a C to G substitution at nucleotide position 431, causing the serine (S) at amino acid position 144 to be replaced by a cysteine (C). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -
