This sequence change replaces isoleucine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 418 of the AKAP9 protein (p.Ile418Arg). This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with AKAP9-related conditions. ClinVar contains an entry for this variant (Variation ID: 1761316). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. The p.I418R variant (also known as c.1253T>G), located in coding exon 8 of the AKAP9 gene, results from a T to G substitution at nucleotide position 1253. The isoleucine at codon 418 is replaced by arginine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
