Variant summary: PLEC c.6329C>T (p.Ala2110Val) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00033 in 149434 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in PLEC causing PLEC-Related Disorders, allowing no conclusion about variant significance. c.6329C>T has been reported in the literature in at-least one individual affected with Slow progressive congenital myopathy (example: Al-Dewik_2019). These report(s) do not provide unequivocal conclusions about association of the variant with PLEC-Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 30919572). ClinVar contains an entry for this variant (Variation ID: 287223). Based on the evidence outlined above, the variant was classified as uncertain significance.

This variant along with a second variant in this gene has been reported in an individual with limb-girdle muscular dystrophy (Al-Dewik et al. 2019. PubMed ID: 30919572). This variant is reported in 0.28% of alleles in individuals of Ashkenazi Jewish descent in gnomAD. At this time, the clinical significance of this variant is uncertain due to the absence of conclusive functional and genetic evidence.
