Member of the FGF family possess broad mitogenic and cell survival activities, and are involved in a variety of biological processes including embryonic development cell growth, morphogenesis, tissue repair, tumor growth and invasion. This protein is expressed during embryogenesis and in the adult cerebellum and cortex and may be essential for vascular growth and normal brain development. This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 187 of the FGF17 protein (p.Asn187Ser). This variant is present in population databases (rs398123026, gnomAD 0.003%). This missense change has been observed in individual(s) with Kallmann syndrome (PMID: 23643382). ClinVar contains an entry for this variant (Variation ID: 50860). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. Experimental studies have shown that this missense change does not substantially affect FGF17 function (PMID: 23643382).
