This sequence change replaces leucine, which is neutral and non-polar, with valine, which is neutral and non-polar, at codon 414 of the WRN protein (p.Leu414Val). RNA analysis indicates that this missense change induces altered splicing and likely results in the loss of 10 amino acid residue(s), but is expected to preserve the integrity of the reading-frame. This variant is present in population databases (rs587778755, gnomAD 0.02%). This variant has not been reported in the literature in individuals affected with WRN-related conditions. ClinVar contains an entry for this variant (Variation ID: 135450). An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Benign". The valine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. Studies have shown that this missense change results in the activation of a cryptic splice site in exon 9 (internal data). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
