The encoded protein plays a role in cell-cycle progression and spindle organization, regulates cytokinesis, interacts with Nephrocystin 8 and is required for cilia formation. Mutations in this gene result in primary cilia abnormalities and classical Joubert syndrome. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. The c.1660C>A (p.H554N) alteration is located in exon 13 (coding exon 13) of the CSPP1 gene. This alteration results from a C to A substitution at nucleotide position 1660, causing the histidine (H) at amino acid position 554 to be replaced by an asparagine (N). Based on insufficient or conflicting evidence, the clinical significance of this alteration remains unclear. -

This sequence change replaces histidine, which is basic and polar, with asparagine, which is neutral and polar, at codon 554 of the CSPP1 protein (p.His554Asn).
