ClinVar contains an entry for this variant (Variation ID: 530728). This variant has not been reported in the literature in individuals affected with NBN-related conditions. This variant is not present in population databases (gnomAD no frequency). This sequence change replaces serine, which is neutral and polar, with leucine, which is neutral and non-polar, at codon 633 of the NBN protein (p.Ser633Leu). RNA analysis indicates that this missense change induces altered splicing and likely results in a shortened protein product. - |

In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
