This pathogenic variant destroys the canonical splice acceptor site in intron 3, and is expected to cause abnormal gene splicing. The c.170-2 A>G variant is not observed in large population cohorts (Lek et al., 2016). Therefore, the presence of c.170-2 A>G is consistent with the diagnosis of an STXBP1-related disorder in this individual. -
