The p.G514S variant (also known as c.1540G>A), located in coding exon 12 of the COL5A1 gene, results from a G to A substitution at nucleotide position 1540. The glycine at codon 514 is replaced by serine, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with classical Ehlers-Danlos syndrome (EDS) (Richer J et al. Arterioscler Thromb Vasc Biol, 2020 Nov;40:2686-2699). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the majority of available evidence to date, this variant is likely to be pathogenic. -

This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 514 of the COL5A1 protein (p.Gly514Ser). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
