Variant summary
Our verdict is Likely benign. The variant received -2 ACMG points: 2P and 4B. PM2BP4_Strong
The NM_017617.5(NOTCH1):c.7639A>C(p.Ile2547Leu) variant causes a missense change involving the alteration of a non-conserved nucleotide. The variant allele was found at a frequency of 0.000000687 in 1,455,812 control chromosomes in the GnomAD database, with no homozygous occurrence. In-silico tool predicts a benign outcome for this variant. 17/21 in silico tools predict a benign outcome for this variant. Variant has been reported in ClinVar as Uncertain significance (★). Synonymous variant affecting the same amino acid position (i.e. I2547I) has been classified as Likely benign.

Familial thoracic aortic aneurysm and aortic dissection Uncertain:1
The p.I2547L variant (also known as c.7639A>C), located in coding exon 34 of the NOTCH1 gene, results from an A to C substitution at nucleotide position 7639. The isoleucine at codon 2547 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear. -
