This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has been identified in 1/246256 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance. - |

The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.416G>A has been reported in the literature in individuals affected with Colorectal Cancer (Fujita_2020), but it has also been reported in multiple unaffected controls (McWilliams_2018, Fujita_2020). These reports do not provide unequivocal conclusions about association of the variant with Cutaneous Malignant Melanoma. Experimental evidence demonstrated the variant disrupts interactions with other proteins/transcription factors (Sun_2010, Al-Khalaf_2013, Al-Khalaf_2017), however, no convincing conclusions about the variant effect could be drawn.
