Variant summary
Our verdict is Uncertain significance. The variant received 0 ACMG points: 2P and 2B. PM2BP4_Moderate
The NM_058195.4(CDKN2A):c.192A>G(p.Pro64Pro) variant causes a splice region, synonymous change involving the alteration of a non-conserved nucleotide. The variant was absent in control chromosomes in GnomAD project. In-silico tool predicts a benign outcome for this variant. 2/3 splice prediction tools predict no significant impact on normal splicing. Variant has been reported in ClinVar as Uncertain significance (★★). Synonymous variant affecting the same amino acid position (i.e. P64P) has been classified as Uncertain significance.

Hereditary cancer-predisposing syndrome Uncertain:1
The c.192A>G variant (also known as p.P64P), located in coding exon 1 of the CDKN2A (p14ARF) gene, results from an A to G substitution at nucleotide position 192. This nucleotide substitution does not change the amino acid at codon 64. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing; however, direct evidence is insufficient at this time (Ambry internal data). Based on the available evidence, the clinical significance of this variant remains unclear. -
Familial melanoma Uncertain:1
