Genes affected
GRHPR (HGNC:4570): (glyoxylate and hydroxypyruvate reductase) This gene encodes an enzyme with hydroxypyruvate reductase, glyoxylate reductase, and D-glycerate dehydrogenase enzymatic activities. The enzyme has widespread tissue expression and has a role in metabolism. Type II hyperoxaluria is caused by mutations in this gene. [provided by RefSeq, Jul 2008]

Primary hyperoxaluria, type II Pathogenic:1

not provided Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Aug 03, 2023 | ClinVar contains an entry for this variant (Variation ID: 204239). This missense change has been observed in individual(s) with primary hyperoxaluria type II (PMID: 11030416). This variant is present in population databases (rs180177325, gnomAD 0.01%). This sequence change replaces methionine, which is neutral and non-polar, with arginine, which is basic and polar, at codon 322 of the GRHPR protein (p.Met322Arg). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on GRHPR protein function. In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. This variant disrupts the p.Met322 amino acid residue in GRHPR. Other variant(s) that disrupt this residue have been observed in individuals with GRHPR-related conditions (PMID: 11030416, 25629080), which suggests that this may be a clinically significant amino acid residue. - |
