The glycine at codon 839 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been detected in two siblings with lower extremity-predominant spinal muscular atrophy 2 (SMALED2) and a family history of relatives with similar clinical features (Rudnik-Schöneborn S et al. Muscle Nerve, 2016 09;54:496-500). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. - |
