This sequence change replaces isoleucine with threonine at codon 523 of the FANCC protein (p.Ile523Thr). The isoleucine residue is weakly conserved and there is a moderate physicochemical difference between isoleucine and threonine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with FANCC-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The threonine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. -

The FANCC p.Ile523Thr variant was not identified in the literature nor was it identified in the dbSNP, or LOVD 3.0, databases. The variant was only identified in ClinVar (classified as uncertain significance by Invitae). The variant was not identified in the following control databases: the Exome Aggregation Consortium (August 8th 2016), or the Genome Aggregation Database (Feb 27, 2017). The p.Ile523 residue is not conserved in mammals and four out of five computational analyses (PolyPhen-2, SIFT, AlignGVGD, BLOSUM, MutationTaster) do not suggest a high likelihood of impact to the protein; however, this information is not predictive enough to rule out pathogenicity. The p.I523T variant (also known as c.1568T>C), located in coding exon 14 of the FANCC gene, results from a T to C substitution at nucleotide position 1568. The isoleucine at codon 523 is replaced by threonine, an amino acid with similar properties. This alteration has been reported in individuals who underwent hereditary breast and ovarian cancer syndrome testing based on personal and/or family history (Thompson ER et al. PLoS Genet, 2012 Sep;8:e1002894; Lerner-Ellis J et al. J Cancer Res Clin Oncol, 2021 Mar;147:871-879). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear. -
