To our knowledge, this variant has not been reported in individuals with nevoid basal cell carcinoma syndrome. The p.F1068L variant (also known as c.3204C>A), located in coding exon 19 of the PTCH1 gene, results from a C to A substitution at nucleotide position 3204. The phenylalanine at codon 1068 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
