Functional assays in this same paper found that the p.Arg217Cys substitution causes a decrease in protein activity (Wimplinger et al. 2006. PubMed ID: 17033964). This variant has not been reported in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. This variant is interpreted as likely pathogenic. -
