The encoded protein is involved in remodeling the cytoskeleton to effect changes in cell shape and migration. This protein interacts with integrins, transmembrane receptor complexes, and second messengers. Defects in this gene are a cause of several syndromes, including periventricular nodular heterotopias (PVNH1, PVNH4), otopalatodigital syndromes (OPD1, OPD2), frontometaphyseal dysplasia (FMD), Melnick-Needles syndrome (MNS), and X-linked congenital idiopathic intestinal pseudoobstruction (CIIPX). This alteration results from a G to A substitution at nucleotide position 7561, causing the glutamic acid (E) at amino acid position 2521 to be replaced by a lysine (K). Based on data from gnomAD, the A allele has an overall frequency of 0.001% (1/181458) total alleles studied. The highest observed frequency was 0.004% (1/27366) of Latino alleles. This amino acid position is highly conserved in available vertebrate species. This alteration is predicted to be deleterious by in silico analysis.
