The encoded protein is involved in remodeling the cytoskeleton to effect changes in cell shape and migration. This protein interacts with integrins, transmembrane receptor complexes, and second messengers. Defects in this gene are a cause of several syndromes, including periventricular nodular heterotopias (PVNH1, PVNH4), otopalatodigital syndromes (OPD1, OPD2), frontometaphyseal dysplasia (FMD), Melnick-Needles syndrome (MNS), and X-linked congenital idiopathic intestinal pseudoobstruction (CIIPX). This variant was previously reported in the SNPDatabase as rs199652065. In the ESP, this variant was not observed in 6273 samples (12546 alleles) with coverage at this position. This nucleotide position is well conserved in available vertebrate species. BDGP and ESEfinder splice site prediction tools do not produce a reliable prediction for the nearby native splice acceptor site. Using the Human Splicing Finder (HSF) splice site prediction tool, this alteration does not have any significant effect on the native splice acceptor site; however, direct evidence is unavailable. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear. -
