Mammalian complex I is located at the mitochondrial inner membrane. This protein has NADH dehydrogenase activity and oxidoreductase activity. It transfers electrons from NADH to ubiquinone. This variant is located in the last exon of NDUFB11; therefore, it is unknown if the resulting mRNA would result in nonsense-mediated decay. However, at least one other protein-truncating variant was described on the last exon in an individual who presented with NDUFB11-related disease (van Rahden et al. 2015. PubMed ID: 25772934). To our knowledge, this variant has not been reported in the literature or in a large population database (http://gnomad.broadinstitute.org), indicating this variant is rare. At PreventionGenetics, we previously detected this variant as de novo in an individual who presented with histiocytoid cardiomyopathy. Note that variable clinical features have been described in patients with NDUFB11-related disease (see, for example, Rea et al. 2017. PubMed ID: 28050600; Amate-Garcia et al. 2023. PubMed ID: 36675256). Taken together, we classify this variant as likely pathogenic. -
