The DNA-binding motifs suggest this protein is involved in the regulation of transcription and chromatin remodeling. Mutations in this gene have been associated with X-linked cognitive disability. The Q1020X variant in the KDM5C gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q1020X variant is not observed in large population cohorts (Lek et al., 2016). We interpret Q1020X as a pathogenic variant that is consistent with the clinical features reported in this individual. -
