This variant has not, to our knowledge, been published in the literature as pathogenic or benign. RET Gly885Glu was not observed at a significant allele frequency in large population cohorts (Lek 2016). Since Glycine and Glutamic Acid differ in polarity, charge, size or other properties, this is considered a non-conservative amino acid substitution. Gly885Glu occurs at a position that is conserved across species and is located in the tyrosine kinase and intracellular domains (Garcia-Barcelo 2004). In silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available evidence, it is unclear whether RET Gly885Glu is a pathogenic or benign variant. We consider it to be a variant of uncertain significance. - |

In addition, the in silico prediction for this alteration is inconclusive. Based on data from gnomAD, the frequency for this variant is above the maximum credible frequency for a Hirschsprung disease-causing variant in this gene based on internally established thresholds (Karczewski et al. Nature. 2020 May;581(7809):434-443; Whiffin et al. Genet Med. 2017 10;19:1151-1158). Based on the supporting evidence, the association of this alteration with MEN2 is unknown; however, the association of this alteration with Hirschsprung disease is unlikely. - |
