Tyrosinase-negative oculocutaneous albinism Pathogenic:2
Pathogenic, criteria provided, single submitter | clinical testing | Center of Genomic medicine, Geneva, University Hospital of Geneva | Jan 08, 2018 | This heterozygous missense variant in the TYR gene was identified in a very young patient with oculocutaneous albinism (type 1A) in combination with another variant (deletion ) in the same gene (compound heterozygosity). This variant was inherited form the mother (asymptomatic), while the second variant was inherited from the father (asymptomatic). - |

not provided Pathogenic:1
Likely pathogenic, criteria provided, single submitter | clinical testing | Labcorp Genetics (formerly Invitae), Labcorp | Sep 28, 2023 | This sequence change replaces tyrosine, which is neutral and polar, with cysteine, which is neutral and slightly polar, at codon 327 of the TYR protein (p.Tyr327Cys). This variant is present in population databases (no rsID available, gnomAD 0.02%). This missense change has been observed in individuals with clinical features of oculocutaneous albinism (PMID: 13680365; Invitae; http://www.didac.ehu.es/antropo/7/7-6/Aquaron.htm). ClinVar contains an entry for this variant (Variation ID: 585280). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt TYR protein function. Experimental studies have shown that this missense change affects TYR function (PMID: 27537549). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic. - |
TYR-related disorder Pathogenic:1
Likely pathogenic, no assertion criteria provided | clinical testing | PreventionGenetics, part of Exact Sciences | Aug 01, 2024 | The TYR c.980A>G variant is predicted to result in the amino acid substitution p.Tyr327Cys. This variant has been reported in individuals with oculocutaneous albinism (King et al. 2003. PubMed ID: 13680365; Loftus et al. 2023. PubMed ID: 37327787). Additionally, this variant has been detected along with a pathogenic TYR variant in multiple individuals undergoing testing for oculocutaneous albinism (PreventionGenetics, internal data). The p.Tyr327Cys substitution is predicted damaging to the protein and functional studies have confirmed it causes a complete ablation of enzyme activity (Mondal et al. 2016. PubMed ID: 27537549). This variant is reported in 0.023% of alleles in individuals of African descent in gnomAD. This variant is interpreted as likely pathogenic. - |
Oculocutaneous albinism Uncertain:1
Uncertain significance, criteria provided, single submitter | clinical testing | Illumina Laboratory Services, Illumina | Apr 28, 2017 | This variant was observed as part of a predisposition screen in an ostensibly healthy population. A literature search was performed for the gene, cDNA change, and amino acid change (where applicable). Publications were found based on this search. However, the evidence from the literature, in combination with allele frequency data from public databases where available, was not sufficient to rule this variant in or out of causing disease. Therefore, this variant is classified as a variant of unknown significance. - |
