LAMA2 (HGNC:6482): (laminin subunit alpha 2) Laminin, an extracellular protein, is a major component of the basement membrane. It is thought to mediate the attachment, migration, and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. It is composed of three subunits, alpha, beta, and gamma, which are bound to each other by disulfide bonds into a cross-shaped molecule. This gene encodes the alpha 2 chain, which constitutes one of the subunits of laminin 2 (merosin) and laminin 4 (s-merosin). Mutations in this gene have been identified as the cause of congenital merosin-deficient muscular dystrophy. Two transcript variants encoding different proteins have been found for this gene. Several computational tools predict a significant impact on normal splicing: Four predict the variant abolishes a canonical 5' splicing donor site. At least one publication reports experimental evidence confirming that this variant affects mRNA splicing (Liang_2017). The variant allele was found at a frequency of 1.3e-05 in 150988 control chromosomes (gnomAD v.3.1.2). c.4311G>A has been reported in the literature in at least one compound heterozygous individual affected with Congenital Muscular Dystrophy (Liang_2017). These data do not allow any conclusion about variant significance. The following publication has been ascertained in the context of this evaluation (PMID: 28182637). One ClinVar submitter has assessed the variant since 2014, and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as likely pathogenic. -

This variant is present in population databases (no rsID available, gnomAD 0.007%). This variant has been observed in individual(s) with congenital muscular dystrophy (PMID: 28182637). ClinVar contains an entry for this variant (Variation ID: 477472). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Studies have shown that this variant is associated with altered splicing, but the impact on the resulting protein product is unknown (PMID: 28182637).
