The tryptophan at codon 347 is replaced by cysteine, an amino acid with highly dissimilar properties. This alteration has been reported in a family with cutaneous melanoma and was found to segregate with disease in 5 of the 6 melanoma cases that were available for testing, including two individuals with multiple primary melanomas, and a case that developed three different primary cancers: melanoma, renal cell carcinoma and prostate cancer (Aoude LG et al. Fam. Cancer 2015 Dec; 14(4):621-8). This amino acid position is highly conserved in available vertebrate species.In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear. - |
