For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects RHO function (PMID: 18987202, 27458239). Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). ClinVar contains an entry for this variant (Variation ID: 13045). This missense change has been observed in individuals with autosomal dominant RHO-related conditions (PMID: 7846071, 33669941). It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (ExAC no frequency). This sequence change replaces glycine with aspartic acid at codon 90 of the RHO protein (p.Gly90Asp). The glycine residue is highly conserved and there is a moderate physicochemical difference between glycine and aspartic acid. -
