The aspartic acid at codon 418 is replaced by tyrosine, an amino acid with highly dissimilar properties. This alteration has been observed in multiple individuals with a personal history that is consistent with MEN1-related disease (Ozturk M et al. J Endocrinol Invest. 2006 Jun;29(6):523-7; Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the supporting evidence, this alteration is interpreted as a disease-causing mutation. -
