The variant allele was found at a frequency of 3.2e-05 in 251468 control chromosomes (gnomAD). c.305C>T has been reported in the literature in at least compound heterozygous individual affected with Congenital Disorder Of Glycosylation Type 1B (Jaeken_1998). These data do not allow any conclusion about variant significance. When the variant was introduced into the mouse ortholog of the MPI gene and expressed in an mpi-null yeast strain, the variant showed 0.6% residual activity (He_2014), indicating loss of function.
