Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251282 control chromosomes. c.442G>A has been observed in compound heterozygous and homozygous individuals affected with Hyperimmunoglobulin D with periodic fever (Houten_2001, D'Osualdo_2005, Bader-Meunier_2011). These data indicate that the variant is likely to be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publications have been ascertained in the context of this evaluation (PMID: 21708801, 15536479, 11313768). ClinVar contains an entry for this variant (Variation ID: 97592).
