The p.Y225* pathogenic mutation (also known as c.675T>A), located in coding exon 7 of the PTEN gene, results from a T to A substitution at nucleotide position 675. This changes the amino acid from a tyrosine to a stop codon within coding exon 7. This mutation has been reported as likely de novo in an individual with clinical features of PTEN-related disease (Steffann J et al. Eur J Hum Genet, 2014 May;22:711-2). In addition, this alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation. -
